Diversity of fungal pathogens associated with loquat and development of novel virulence scales2021 •
Loquat [Eriobotrya japonica (Thunb.) Lindl.] is an important fruit crop in Pakistan; however, a constant decline in its production is noted due biotic and abiotic stresses, particularly disease infestation. Fungal pathogens are the major disease-causing agents; therefore, their identification is necessary for devising management options. This study explored Taxila, Wah-Cantt, Tret, Chatar, Murree, Kalar-Kahar, Choa-Saidan-Shah and Khan-Pur districts in the Punjab and Khyber Paktoon Khawa (KPK) provinces of Pakistan to explore the diversity of fungal pathogens associated with loquat. The samples were collected from these districts and their microscopic characterizations were accomplished for reliable identification. Characterization and Pathogenicity of Fusicladium eriobotryae, the Fungal Pathogen Responsible for Loquat Scab2009 •
To characterize the fungal pathogen responsible for loquat scab and establish differences in pathogenicity in loquat, eight strains identified as Fusicladium eriobotryae were isolated from either loquat leaves or fruit showing scab symptoms in Spain. Loquat plants belonging to the cv. Peluche were infected via a newly developed infection system that was based on spraying susceptible loquat plants with fungal spore suspensions, keeping the plants for 1 week in 100% humidity, and then transferring the plants to ambient relative humidity in greenhouses. Scab symptoms were analyzed and pathogenic characterization of all F. eriobotryae strains revealed different degrees of aggressiveness. Based on infection progression and severity of scab symptoms, strain ST1 was confirmed as the most aggressive in cultivars in the Mediterranean region. Alternaria alternata is a fungus which has been recorded causing leaf spot and other diseases on over 380 host species of plant. It is an opportunistic pathogen on numerous hosts causing leaf spots, rots and blights on many plant parts. A leaf spot and fruit rot on loquat, caused by A. alternata, was found in the plantations and entertainment venues for the first time in Iran in Sari city, Mazandaran province. The fungus was isolated from leaves and fruits, and its pathogenicity was confirmed. Pathogenicity tests showed that A. alternata could infect loquat, which developed the same symptoms after inoculation as those observed naturally in the field. The fungus was identified based on morphological characteristics and confirmed using comparisons of DNA sequences of Alt α 1 locus. This report is the first on loquat leaf spot and fruit rot disease caused by A. alternata in Iran.

In Pakistan, loquat (Eriobotrya japonica) in an emerging fruit and provides well economic return to growers. Percentage disease incidence of necrotic leaf spot (Alternaria mali) ranged from 25 to 46.16% in Taxila, Wahcant, Khanpur, Kalar Kahar, Choa Saiden Shan, Tret, Chatar and Murree. 38 isolates were characterized for cultural, morphological and pathogenic variations. Light to dark olivacious colonies were appraised, cottony to velvety with regular to irregular margins and variations in size and shape of conidia were observed between isolates. Length of conidiophores was ranging from 24.6 to 68.5 µm and nine was the highest septation in isolate ALT7WT1. Nucleotide sequences of inter transcribed spacer region (ITS1, 5.8S, ITS2) from 4 highly virulent isolates were submitted in the database of NCBI under accession numbersKR232489, KR232490, KT154010 and KT154011 respectively. Evolutionary history was computed with available sequences of A. mali, A. lternata, A. zinnae and A. tenuissima which were reported from Pakistan, USA, Sweden, Newzeland and India. Evolutionary tree showed4 different sub-trees and submitted sequences were observed in A. mali sub-tree. Amino acid sequences were exhibiting 100 to 99% genetic homology with A. mali isolate (FCBP1343) reported form Pakistan. Amino acid sequences were compared with FCBP1343 and differences at position number 18 (valine was replaced with glutamate), 57 (glutamine with leucine), 84 (alanine with glycine), 163 (glutamine instead of histidine) and 171 (serine was replaced with phenylalanine) were observed. Cultural, morphological, pathogenic and molecular characterization of A. mali associated with necrotic leaf spot of loquat was recorded first time in Pakistan.

Novel fungal genera and species associated with the sooty blotch and flyspeck complex on apple in China and the USA2010 •
Fungi in the sooty blotch and flyspeck (SBFS) complex cause blemishes on apple and pear fruit that result in economic losses for growers. The SBFS fungi colonise the epicuticular wax layer of pomaceous fruit but do not invade the cuticle. Fungi causing fuliginous and punctate mycelial types on apple are particularly difficult to identify based on morphological criteria because many species in the SBFS complex share the same mycelial phenotypes. We compared the morphology and nuclear ribosomal DNA phylogeny (ITS, LSU) of 11 fungal strains isolated from SBFS blemishes on apple obtained from two provinces in China and five states in the USA. Parsimony analysis, supported by cultural characteristics and morphology in vitro, provided support to delimit the isolates into three novel genera, representing five new species. Phaeothecoidiella, with two species, P. missouriensis and P. illinoisensis, is introduced as a new genus with pigmented endoconidia in the Dothideomycetes. Houjia (Capnodiales) is introduced for H. pomigena and H. yanglingensis. Although morphologically similar to Stanjehughesia (Chaetosphaeriaceae), Houjia is distinct in having solitary conidiogenous cells. Sporidesmajora (Capnodiales), based on S. pennsylvaniensis, is distinguished from Sporidesmium (Sordariomycetes) in having long, multiseptate conidiophores that frequently have a subconical, darkly pigmented apical cell, and very long, multi-euseptate conidia.
