Ducrosia Anethifolia is a threatened medicinal herb belongs to the family Apiaceae. This important medicinal plant is a native medicinal plant in Iran and has a restricted distribution in the world. It is used to treat of headache and backache in folk medicine. To callus induction, Leaf and lateral bud were used as explants and the Culture media was MS medium supplemented with 3% w/v sucrose, 0.7% w/v agar and 1gl -1 myoinositol at various concentration of cytokinins and auxins. After Five weeks, the calluses were transferred into regeneration medium that included MS medium supplemented with the combination of BA(0.5, 1 and 2 m gl -1 ) and NAA(0, and 0.5 mgl -1 ). Costus igneus is an important medicinal herb which is known to reduce the levels of insulin in the case of Diabetes mellitus. Efficient protocol for micropropagation using various explants is developed and the observations on different parameters are recorded. The surface sterilization of the explants at various time intervals and varying concentration of 70% ethanol, sodium hypochlorite and mercuric chloride was optimized in order to reduce the overall contamination levels during the study. This plant was effectively micropropagated using various explants with the standardized protocol in both standard MS medium and LS medium at various hormone levels. The leaf, node, internode, rhizome were the explants used for the study. Callus was induced on the 4 th d in MS medium and on the 3 rd d in LS medium at the hormone concentration of BAP (0.4 mg/L) + KIN (0.2 mg/L) + NAA (0.1 mg/L) + IAA (0.2 mg/L) + IBA (0.2 mg/L). Shoot regeneration using the nodal and internodal explants was observed in MS and LS medium with BAP (0.4 mg/L) + KIN (0.25 mg/L) + NAA (0.1 mg/L) + IAA (0.2 mg/L). Qualitative and quantitative phytochemical analysis of hexane, ethyl acetate, diethyl ether, acetone and methanol extracts of wild plant and callus were carried out with various reagents. Methanolic extract was found to have more total phenolic and flavonoid content in wild plant and callus.
Objective: To study callus induction from different explants (internode, leaf, root) and in vitro plantlets propagation from medicinally important plant Achyranthes aspera L. Methods: Sterilized explants were prepared by using 0.1% HgCl2 and 0.5% Bavistin and callus was obtained when cultured onto Murashige Skoog’s (MS) medium by using different concentrations and combination of 2,4-D, NAA, BAP, IAA, IBA with 3% sucrose and 0.8% agar. Induced callus was immediately transferred to MS medium containing at different concentrations of phytohormones for shootlets and rootlets induction respectively. Results: Sterilization treatment of 0.1% HgCl2 for 2-3 min and Bavistin 0.5% for 10-12 min showed the highest percentage of asepsis and survival rate. Maximum induction of callus was obtained from a combination of 2.0 mg/L 2,4-D and 0.5 mg/L NAA from leaf. Highest shootlets number (4.83依0.17) and length (3.8依0.16) cm were observed on full strength MS medium when fortified with BAP 4.0 mg/L and KIN 0.5 mg/L. Concerted efforts of BAP 2.0 mg/L and NAA 0.5 mg/L on full strength MS medium showed highest leaf number (6.77依0.94). In vitro raised shoots were allowed to root on different strengths of MS medium fortified with IAA and IBA at different concentrations. Experimentally, 3.0 mg/L IBA was enabled to induce maximum rootlets number (10.0依9.82) on full strength MS medium. Afterwards, regenerated shoots with well developed roots were successfully subjected to hardening process and were acclimatized. The survived plantlets showed 66.67% survival frequency without any morphological abnormality. Conclusions: The results demonstrated that different explants were good source of callus induction, morphology analysis as well as indirect plantlets regeneration.

In Vitro Callus Induction and Regeneration of Medicinal Plant Datura innoxia2019 •

Devegulu hollyhock is used by public for the treatment of certain diseases. This study investigated the appropriate protocols for in vitro callus formation, regeneration and micropropagation of various explants of 14-day and 28-day old Devegulu seedlings germinated from the seeds in a sterile environment. Different concentrations of 2,4- dichlorophenoxy acetic acid (2,4-D) were used as plant growth regulator and Murashige and Skoog basal medium was used as basic medium. Regenerated plantlets were firstly transferred to rooting medium to promote root formation, after that they were planted into soil. Both 14-day and 28-day explants produced a higher rate of callus in MS medium including 2,4-D auxin hormone. The regenerated plantlets were obtained from the shoot tip and node explants of 28 days old seedlings after incubation in MS medium for 6 weeks. Orthosiphun stamineus is a herbaceous plant that is popularly known as Misai Kucing. It is widely used in traditional medicine as diuretic agent. This study was divided into two parts that was the in vitro production of complete plantlet through axillary branching and callus culture derived from leaf explant. In axillary branching method, sterilization was conducted using 0.02mg/100ml of mercuric chloride followed by rinsing with 20% and 50% of Clorox for 20 minutes and 5 minutes respectively. This sterilization method was able to remove the contaminants from the surface of the axillary stem and almost 70% of the explants were survived. Axillary bud was placed on Murashige and Skoog (MS) basic medium and cultured for 1 month. The in vitro shoot was inoculated on MS medium which was supplemented with different concentrations of BAP and NAA. The medium that contained 1.0mg/L of BAP gave the best shoot multiplication (13.25) and shoot length (6.23cm) after 8 weeks in culture. Root formation in term of percentage of root (70%) and the number of root produced (10.50) were the best when shoot inserted into medium contained 6mg/L IBA after 3 weeks in culture. However, MS medium that was supplemented with 2 mg/L IBA enhanced in the root length (3.85 cm). Meanwhile, in callus culture, the leaf explant was placed on MS medium containing with various concentrations of 2,4-D for induction of callus. The optimum level of callus induction and proliferation rate (0.42) were obtained with 4mg/L 2,4-D. The callus cells were tested in medium with Evan’s Blue staining and the result showed that the cells were embryogenic. However, the shoot induction from the callus was failed in all tested mediums containing different combinations of BAP and 2,4-D.
